They said the winning ticket was sold in a supermarket in the Florida city of Zephyrhills, which has a population of at least 10,000.
It was not clear if anyone had yet come forward to claim the record win.
Forty-two US states, the District of Columbia and the US Virgin Islands participate in the Powerball lottery.
The numbers drawn in the multi-state lottery on Saturday night were 10, 13, 14, 22, 52 , with the Powerball 11.
"This would be the sixth Florida Powerball winner and right now, it's the sole winner of the largest ever Powerball jackpot," Florida lottery official Cindy O'Connell told the Associated Press news agency.
"We're delighted right now that we have the sole winner."
